The manor remained in the hands of the Dukes of Bedford until 1554 when it was sold to Sir Thomas Smythe (d.1591), a prominent London merchant and Lord Mayor. Sir Thomas Smythe built himself a large mansion in 1580, which is now known as Lansdowne House.

The town of Harpenden developed around the Manor House and by the early 18th century it had a population of around 700. The coming of the railways in the 19th century led to a further increase in population and the town became a commuter town for London.
