The city was located on an important Roman road (now called Watling Street), which connected London with Winchester. In the late Saxon period, Twyford was a royal manor belonging to the king of Wessex.

During the Norman Conquest, the city was given to the Bishop of Winchester. Twyford remained under ecclesiastical control until the 18th century.

The city was an important stopover for pilgrims travelling to the shrine of Thomas Becket at Canterbury. In the 14th century, the city was granted a charter by King Edward III, which allowed it to hold a weekly market and an annual fair.

During the English Civil War, Twyford was a Parliamentarian garrison town. In 1644, a Royalist army under the command of Sir Ralph Hopton attacked and captured the city.

In the 18th century, the city became a coaching stop on the London to Exeter stagecoach route. The construction of the London and Southampton Railway in the 1830s led to a decline in coaching traffic.
